Pharma automation and 21 CFR Part 11

In pharmaceutical manufacturing, automation has to control the process and prove it. We build both in.

Freeze dryer SCADA exported worldwide

Since about 2005 we have developed the control and monitoring software for lyophilizers built by Lyophilization Systems Inc. (New York) and its Indian arm. The machines are exported to Southeast Asia, the Middle East, Europe, China and Russia, and we commissioned two systems on site in the United States. The software runs the full recipe (freezing, primary drying, secondary drying), controlling shelf temperature, chamber pressure, condenser temperature and vacuum, with every parameter and operator action recorded to 21 CFR Part 11.

API and formulation plants

  • Dr Reddy's Laboratories: reactor automation for an API process on Honeywell PLC.
  • Aurobindo Pharma: HVAC automation on Honeywell HC900 to hold cleanroom conditions.
  • Piramal: batch process automation for an API.
  • Gland Pharma and other sites: annual maintenance contracts and iFIX support.

What Part 11 automation includes

User management with role-based access, electronic signatures on critical actions, audit trails that cannot be edited, secure historian storage, batch reports generated from recorded data, and documentation to support validation (URS, FDS, IQ, OQ).

For project heads and buyers

The batch record writes itself. Every setpoint change and every operator action is logged with a name and time, and nobody can edit the log.

Why it matters

FDA, EU and CDSCO inspectors ask for the audit trail. A system without Part 11 features cannot produce one.

What you receive
  • Part 11 configured iFIX with roles, e-signatures and audit trail
  • Batch reports from recorded data
  • Validation support: FDS, IQ, OQ documents
  • AMC with controlled change management
Helpful to have
  • Process description and recipe parameters
  • User roles and signature points
  • Your validation plan or QA template
Typical duration

Three to five months including validation documentation.

How a project runs

  1. Scope callWe go through your P&ID, I/O list or tender clause and agree what the control system has to do.
  2. Technical offerA control philosophy, architecture drawing, bill of materials and price. No surprises later.
  3. Engineering and buildDrawings for approval, PLC logic and SCADA screens built, panels assembled and FAT tested with you present.
  4. CommissioningInstallation supervision, loop checks, start-up and operator training on your site.
  5. SupportDocumentation handover and an annual maintenance contract with a named engineer.

What to send us for a quote

Attach these to your enquiry. The three marked essential are enough for us to start a budget.

  • Process description, P&ID and recipe parametersEssential

They define what is recorded, which setpoints are critical and which actions need a signature.

  • URS, or your QA's URS template, and the validation master plan if one existsEssential

Validation traces back to the URS, so we need it first.

  • User roles and signature pointsEssential

Who may change, approve and release is configured into the system, not added later.

  • Existing SCADA and PLC versions and licences, if upgrading

Part 11 controls depend on the SCADA version and its security configuration.

  • Your QA's templates and document numbering for FDS, IQ and OQ

We write to your formats so QA does not have to rewrite them.

  • Markets you supply: US FDA, EU, CDSCO or others

It decides which inspectors read the audit trail and how it must be reviewed.

  • Backup, archive and data retention policy

Audit trails and batch records must stay readable for the whole retention period.

  • Instrument list with calibration status

Qualification runs on calibrated instruments only, so gaps delay OQ.

Do not have all of it? Send what you have; we work from a site visit for the rest.

What drives cost and schedule

Validation documentation depth
A full IQ and OQ with traceability to the URS takes far more days than a short protocol.
Number of roles and signature points
Each one is configured, tested and written into the documents.
Recipe and batch complexity
More phases and parameters mean more screens, reports and test cases.
Upgrading an existing system
Existing data and screens must be migrated without breaking the audit trail.
Your QA review cycles
Each document round with QA adds days, and unclear comments add more.
Archive and review reporting
Audit trail review reports and long-term archive need extra configuration.

Typical timeline

  1. Scope, URS and validation plan2 to 3 weeksRequirements and validation approach agreed with QA.
  2. FDS and design approval2 to 4 weeksFunctional design written and approved by QA.
  3. Build and configure4 to 6 weeksRoles, signatures, audit trail, screens and reports configured.
  4. FAT and IQ1 to 2 weeksInstallation qualification documents and factory tests.
  5. Site installation and OQ2 to 4 weeksOperational qualification executed on site.
  6. Training and handover2 to 3 weeksValidation pack handed over; PQ stays with your QA.

Overall: Three to five months including validation documentation.

Budget

Every plant is different, so we price from the I/O list and scope rather than a rate card. Send the documents above and you will have a budget figure within a week, and a fixed price after the scope call.

Common questions

What does 21 CFR Part 11 require from a SCADA system?

Secure, time-stamped electronic records, audit trails of every change and operator action, unique logins with electronic signatures, and protection of records for their retention period.

Why GE iFIX for Part 11?

iFIX provides electronic signatures, audit trails and secure historian data, and is widely accepted in regulated pharmaceutical plants. Our freeze dryer SCADA has been built on it for about twenty years.
